3.7. Cell viability
Cell viability was detected by MTT assay which relies on mitochondrial
metabolic capacity of viable cells. The results showed,
that water extracts had cytotoxity at 0.2 mg/mL (Fig. 1). After
treatment with t-BHP (80 lM) for 24 h, the cell viability was
41 ± 3.79% of control, and decreased to 19 ± 0.77% (at 0.1 mg/mL),
15 ± 0.57% (at 0.05 mg/mL) and 14 ± 0.73% (at 0.025 mg/mL) whenthe cells were pretreated with TJP water extract for 1 h (Fig. 2).
Interestingly, these results showed pretreatment with TJP water
extracts with t-BHP cause an increase in cytotoxicity. TJP water extracts
showed very strong radical scavenging activity, but water
extracts some contents may be acted toxicity on the contrary.
However, we found that cell viability increased to 63 ± 0.86%,
78 ± 2.42% and 82 ± 3.24% at 0.025, 0.05 and 0.1 mg/mL respectively
when the cells were pretreated with TJP ethanolic extract.
In positive group, pretreatment with NAC increased cell viability
to 77 ± 1.73% at 5 mM. These results demonstrate that the ethanolic
extract of TJP protected Chang cells from t-BHP induced cell
