The effect of increasing substrate concentration on the enzyme reaction rate follows a typical Michaelis–Menten equation with soluble starch as a substrate.
Double reciprocal plots
using least square regression to fit the plots were used to calculate the KM. It should be considered
that the lower Km value of starch is related to the higher affinity for starch compared with other substrates.
The Km and Vmax values of the amylase enzyme were calculated as 2.7 mg/ml and 34.30 units/minute/mg of protein, respectively, at pH 5.0 and at 70 C
