Laboratory measures in the HDM challenge study
Three outcomes were assessed. First, markers of T-cell activation
(CD691HLA-DR1) on CD41 and CD81 T cells and the CD4/CD8 T-cell ratio
(all from peripheral blood) were determined by means of flow cytometry, as
described in the Methods section in this article’s Online Repository. Second,
serum specific IgE (ssIgE) levels for D pteronyssinus were measured by using
a commercial ImmunoCAP assay (Thermo Fisher Scientific, Waltham, Mass)
on samples collected before challenge with HDM in the ACC. Third, nasal brushings
were used to collect nasal cells from M1 participants before and after (from
the contralateral nare) exposures 1 and 4in ACC-I (seetheMethodssectioninthis
article’s Online Repository). Total RNA from nasal cells was isolated for RNA
sequencing, as described in the Methods section in this article’s Online Repository.
Samples from 13 M1 participants met quality control criteria for RNA-Seq
