The leaf tissues were infiltrated with and embedded in epoxy resin (Epon 812) in capsules and then transversely sectioned with a LKB-V-type ultramicrotome with thicknesses of 3 m for light microscopy observation. Sections were stained with periodic acid Schiff’s reagent (PAS) and subsequently observed using an OLYMPUS BX51 microscope (photographed with OLYMPUS DP71 camera system). Thin sections (<100 nm) were stained with uranyl acetate and lead citrate. The sections were then photographed, and chloroplast ultrastructure was observed with transmission electron microscopy (TEM; JEM-1200EX).
