2.2.7 SDS-PAGE Analysis
The SDS-PAGE analysis of sericin was performed
according to Laemmli34. Sample was mixed with an
equal amount of SDS-PAGE sample buffer (0.5M
Tris-HCl of pH 6.8, 2.5% SDS, 5% 2-mercaptoethanol,
10% glycerol) and boiled for 5 min. The resolving gel
was 12% and stacking gel was 5%. The electrophoresis
was carried out at constant voltage of 90V for 4h using
an Enduro electrophoresis system (Labnet International
Inc., USA). After electrophoresis, the gel was stained
with Coomassie Brilliant Blue R-250. After staining the
gels for 30 min, de-staining was carried out. Molecular
weights were estimated using molecular weight marker
(MWM) procured from Genni-India.
