2.3.6. Desulfation of fucoidan
The native and deacetylated fucoidans AmF3 and AmF3DA(100 mg) were desulfated using a modification (Thinh et al.,2013) of the previously described solvolytic desulfation method(Nagasawa, Inoue, & Kamata, 1977). In brief, 10 mL aliquots of thefucoidans (10 mg/mL in water) were passed through an Amber-lite CG-120 column (200–400 mesh, H+-form, Serva, Germany)with H2O. After neutralization with pyridine (0.5 mL), the solutionswere lyophilized. The resulting pyridinium salts of the fucoidanswere dissolved in DMSO (9 mL) and pyridine (0.5 mL), then incu-bated at 100◦C for 3 h. The solutions were exhaustively dialyzedagainst distilled water using a 5 kDa cut-off membrane for 48 h andlyophilized. The yields of desulfated (DS) fucoidans AmF3DS andAmF3DADS were 23.1 and 20.7 mg, respectively.
