2.1.2. Plant material
Internodal cells of Chara corallina were used. The alga was cultured in a large tank containing a layer of pond mud. The tank was illuminated with a 40 W luminescent lamp (day/night rhythm, 14/10 h). One to two days prior to the experiment, giant internodal cells 0.7–0.8 mm in diameter and ~ 60–80 mm long were isolated from neighboring cells and kept in artificial pond water (APW) containing (mM): 0.1 KCl, 1.0 NaCl, 1.0 CaCl2, 1.0 HEPES/NaOH, pH 7.4. Temperature was maintained at 18–20 °C.
