Broodstock were collected in the Strait of Georgia, BC, off Denman
Island in October 2010 and in Kulleet Bay, BC in October 2011, and
had a mean shell length and live weight of 157.7 ± 1.9 cm and
1.28 ± 0.05 kg (±SE, n = 47) and 149.6 ± 2.1 cm and 1.40 ± 0.04 kg
(n=50), respectively. Approximately 25 animalswere laid horizontally
on the bottomof a shallowtank (L ×W× H: 1.2 × 0.9 × 0.3m) and provided
with flow-through, sand-filtered, and UV-treated seawater at
3–4 l min−1 and 8–12 °C. A single-algal diet of CM or TL was drip-fed
at a daily ration of 4–6 × 109 cells ind.−1. Spawning was induced by
adding excessive TL to the holding tank. Fertilized eggs obtained from
multiple parents were hatched in shallow tanks (L × W × H:
1.2 × 0.9 × 0.3 m) at a density of b30 eggs ml−1 and a temperature of
12–15 °C. After 48–60 h, newly developed D-larvae were collected and
used for various experiments or for further rearing to produce juveniles,
