siRNA transfection
siRNA targeting FOXM1and NF-kBp65 was used for the
silencing of FOXM1 and NF-kBp65, respectively in MDA-MB-
231 cells. Scrambled non-target siRNA was used as a negative
control. For siRNA transfections, 2x105cells suspended in 2 ml of
medium were seeded into a 6-well plate. After incubation for 24
hrs, cells were transfected with 50 nM siRNAs by using RNAiMax
(Invitrogen, CA) according to the manufacturer’s instructions.
After 72 hrs proteins were collected and gene silencing efficiency
was measured using immunoblotting. Expression of FOXM1 and
EMT associated markers were checked on the silent proteins.
