Abstract
Natural products from plant are still the alternative source in the search for anticancer drugs that can have a
direct effect on both cancer cell removal and also minimize the side effects to patients. The cytotoxicity
actions of anticancer compounds coupled with the ability to activate anticancer immune responses would
be an ideal strategy to combat the disease. Recently, we have isolated a flavonoid from Brucea javanica
leaves which has shown relatively potent cytotoxic activity in several cancer cell lines. However, it is not
known if this compound has any potential to modulate the activity of immune cells. The flavonoid fraction
from Brucea javanica (BJLM-C8) was derived through bioassay-guided fractionation using methylene
blue assay in several cancer cell lines: CaOV-3, HeLa, MDA-MB 231, HepG2, MG63 and non-cancer cell
lines Vero and L929. The flowcytometric analysis revealed that BJLM-C8 has the ability to induce
apoptosis by increasing the bax, p53 and caspase-3 protein expression. It also decreases the expression of
pro-inflammatory cytokines TNFK and IL-1β in human blood culture that stimulated with 10μg/ml
Lipopolysaccharide. Besides, the IFN
