Destiny Points are a new form of rewards acquired in the final stages of the Ancient Dungeons involving Legendary Heroes, such as Disarm, Black and White, and the Gravity Labyrinth. They are indicated by five heart slots, filling up each time you obtain one.
Each time you clear the stage, there is a small chance of acquiring a single Destiny Point. Buying a Destiny Potion from the shop at the cost of 5 Jewels will double your chances of acquiring one. Once you collected 5 Destiny Points, you are guaranteed to recruit one of the two Legendary Heroes at random in each stage.

A music note emote may indicate no reward. A heart emote indicates a Destiny Point reward. Both of these two emotes have a small chance of recruiting the heroes.A blush emote guarantees a recruitment of one of the heroes.
It is still possible to recruit the Legendary Heroes without collecting 5 Destiny Points, though the chances are just as low. By looking at the emote bubbles above the heroes' heads, you can tell whether you will get a Destiny Point, one of Legendary Heroes, or nothing at all.
Also, even if the time limit for the dungeon had run out, the Destiny Points you accumulated in a particular dungeon will be carried over to the next time that dungeon reopens. The Destiny Points do not carry over to the other dungeons however. For example, if you have 3 Destiny Points in the final stage of The Void and it closes, you will still have 3 Destiny Points when The Void opens again, but not for Path of Rage or Road to Ruin that open next.
