HRM real-time PCR assay
For differential detection, a LightCycler 480 High Resolution Melting Master Kit (Roche Applied Science, Mannheim, Germany) was used. The reaction mixture contained 1× LightCycler 480 HRM Master Mix, which comprises HRM dye (Roche Applied Science), 2.25 mM MgCl2, and each of 0.4 μM Schis_F and Schis_R primers. The total reaction volume was 20 μl. The PCR cycling for HRM curve presentation was done under the following conditions: 1 hold at 95˚C for 10 min; 45 cycles of 95˚C for 10 sec, 55˚C for 8 sec, and 72˚C for 15 sec; then, the mixture was held at 95˚C for 10 sec and 60˚C for 30 sec.
