2.6. Microbiological analyses
The numbering of different microbial populations in brine was
carried out during fermentation. The viable mesophilic counts
were estimated on Plate Count Agar Standard (Oxoid) incubated
at 32 C for 48 h. The lactic acid bacteria were grown on de Man
Rogosa and Sharpe Agar (MRS, Oxoid) charged with 50 mg L1 of
Nystatin (Sigma) at 32 C for 48 h in anaerobic conditions. The population
of yeasts and moulds was estimated on Sabouraud
Chloramphenicol Agar (SAB, Bio-Rad) at 25 C for 48 h. Coliform
bacteria were enumerated in Violet Red Bile Agar (Oxoid) at
35 C for 48 h, Staphylococcus spp. in Mannitol Salt Agar (MSA,
Oxoid) at 32 C for 72 h. The analyses were done in triplicate and
the results expressed as Log CFU mL1.
