Protein concentration was determined at 595 nm using a Dcprotein assay kit purchased from Bio-Rad Laboratories (Hercules,CA, USA) based on the method of Bradford [21]. The total pro-tein concentration of the enzyme solution was estimated from acalibration curve that was constructed using bovine serum albu-min (BSA). Analytical SDS-PAGE was performed using 10% (w/v)acrylamide in gels as described by Laemmli [22]. The protein bandswere visualized with Coomassie Brilliant Blue R-250 (Bio-Rad Lab-oratories, Inc., Hercules, CA, USA) staining. Casein zymographystaining was performed as described elsewhere [6]. The molecu-lar mass of the purified protease was analyzed in the linear modeby MALDI-TOF/MS using a Voyager DE-RP instrument (AppliedBiosystems/PerSeptive Biosystems, Inc., Framingham, MA, USA).Data was collected with a Tektronix TDS 520 numeric oscillographand analyzed using the GRAMS/386 software (Galactic IndustriesCorporation, Salem, NH, USA).
