Klotho may act to protect the cardiovascular system by regulating the NO synthase [66] responsible for generating
endothelium-derived nitric oxide (NO) [66,67], which acts to
regulates vasomotor tone.
Klotho can suppress ageing and oxidative stress by increasing
the expression of peroxiredoxins (Prx-2 and Prx-3) and thioredoxin reductase 1 (Trxrd-1) that act together to reduce ROS
[68]. In Klotho mutant mice, there is a severe decline in cognition and this was associated with an increase in oxidative stress
[69].
