2.5 Microsatellites and genotyping
Eighty-three, fluorescently labeled, microsatellite markers were tested for polymor¬phism in the F, family. Marker details were published in three sources: (1) Crooijmans et al. (1997) for markers with the prefix MFW, (2) Alia et al. (1999) for markers with the prefix CCA (3), and David et al. (2001) for markers with the prefix Koi. Products separation and fragment analysis were carried out as in the work of David et al. (2001) using an ABI 377 DNA sequencer and Genescan software (Perkin Elmer, Foster City, CA, USA). Of the 83 markers tested, 43 were informative, and were used for genotyping the six DNA pools. For each marker, the frequency of each allele in the pool was calculated by dividing its peak area by the total area of all peaks.
