Traditional method. The classical method was used for
preliminary categorisation and identification. The yeast isolates were categorised into groups based on macroscopic and microscopic similarity. The growth characteristics of the yeast colonies on the agar media were the main macroscopic features, whereas the shape,size and arrangements of the spores were the microscopic (Kurtz-man and Fell, 1998; Pitt and Hocking, 1999). Yeasts with similar phenotypic features were set as one group. One hundred one
representative yeast isolates were then selected randomly for each
group to identify the isolates at a species level using molecular techniques.
