Streptomyces inoculum was prepared using ISP2 broth for 48 hours of incubation at 28º C in rotary
shaker. Crude pigment from strain was produced through solid state fermentation using wheat bran and rice
bran medium. About 10% of inoculum was transferred into wheat bran and rice bran in 250 ml flask.
After 14 days of incubation, the crude pigment from both the medium was extracted by solid-liquid
extraction using ethyl acetate and concentrated to dryness using rotary evaporator. Further, the pigment
was separated using readymade silica gel coated on alumina sheets (Hi Media, Mumbai). Separation
was done using various organic solvents such as chloroform, methanol, acetic acid, and n-hexane in
different proportions. After the development of chromatogram, separated pigment was
observed under UV lamp and also by using iodine chamber. After optimizing the solvent system, high quantity
of pigmented was separated with TLC and concentrated.
