2.2. Lychee fermentation and sample collection
Triplicate fermentations by different yeast cultures (T. delbrueckii PRELUDE monoculture, sequential culture, simultaneous
culture and S. cerevisiae MERIT.ferm monoculture) were conducted in 500-mL sterile Erlenmeyer flasks (250 mL of juice for each flask). For monoculture fermentations, 2.5 mL of a T. delbrueckii PRELUDE pre-culture and 2.5 mL of an S. cerevisiae MERIT.ferm pre-culture were inoculated, respectively. For simultaneous fermentation, 2.5 mL of a T. delbrueckii PRELUDE preculture and 2.5 mL of a S. cerevisiae MERIT.ferm pre-culture were inoculated simultaneously. For sequential fermentations, 2.5 mL of a T. delbrueckii PRELUDE pre-culture were inoculated first on day 0, and 2.5 mL of a S. cerevisiae MERIT.ferm pre-culture were inoculated subsequently on day 4. All flasks were fitted with nonabsorbent
cotton wool fitted and covered with aluminum foil to establish semi-aerobic conditions at early stages of fermentations.
Fermentations were carried out statically at 20C for 12 days. Samples were collected at intervals (day 0, day 2, day 4, day 6, day 9, and day 12) and were stored at 20C until analysis.
