Using specific optical filters, three different forms have been detected simultaneously in samples of living bacteria (Heim and Tsien, 1996). Patterson et al. (1997), in comparing properties of five Green fluorescent protein variants relevant to quantitative imaging in live cells, suggested that no single variant was ideal for all application, but each one offers advantages and disadvantages for imaging living cells.
