Previous studies report that the expression of FDH in plants is
induced under conditions of submergence stress in plants (18).
Thus, banana leaves were subjected to submergence stress by
soaking in treatment solution (containing 5 mM KHCO3 and 5 mM
MES) for 0, 2, 8 and 12 h. The tissue was then homogenised in an
isotonic buffer for chloroplast isolation by sucrose density gradient
centrifugation. The soluble protein fraction was extracted from the
intact chloroplasts and used to perform a Western blot analysis
with the anti-FDH antibody described above. The result showed
that the intensity of the FDH signal (Fig. 5C) became slightly weaker
under submergence stress compared to that in the sample without
submergence stress (0 h), indicating that submergence stress might
decreased the expression level of FDH.
