Determination of minimum inhibitory concentration
for antimicrobial activity by agar dilution method7,8
Two fold dilution of the selected fraction from hexane
extract was made by diluting with hexane. 0.5 mLsample in hexane was mixed with 4.5 mL agar media
quickly and thoroughly. The mixture was poured into petri
dish with a diameter of 6.0 cm. Ten types of pathogenic
bacteria were used to test for antibacterial action. These
bacteria included S.aureus ATCC 25923, E. coli ATCC
25922, P. aeruginosa ATCC 27853, Methicillin-resistant
S. aureus (MRSA), Methicillin-sensitive S. aureus (MSSA)
and the isolated strains from patients which were
Acenobacter baumannii, P. aeruginosa, E. coli, E. cloacae
and K. pneumoniae. Three types of pathogenic fungi to
test for antifungal activity were C. albicans, A. flavus and
T. mentagrophytes. The tests were performed in the
same manner as the determination of MIC for
antibacterial activity. Suspensions of micro-organisms
were prepared with turbidity equal that of McFarland No.
0.5. These suspensions were inoculated on surface of
agar plates. Agar test plates were incubated under
optimum condition for each micro-organism.
