2.2. Preparation of media and content analysis
Separate culture media of 7 different types (A–G) were prepared
based on nitrogen free extract (NFE) and crude protein (CP) content
and by mixing 12 raw materials (Table 1) to check the productivity
of fruiting bodies with long shelf life. Each medium containing
5–11 raw materials was analyzed for its components based on previous
report (Van Soest et al., 1991) and the Association of Official
Analytical Chemists (AOAC, 2000). The analysis includes the determination
of amino acid, CP, crude fat, moisture content, dry matter,
ash, crude fiber, acid detergent fiber (ADF), neutral detergent fiber
(NDF), and minerals such as magnesium, calcium, iron, copper, and
manganese. All values were determined on a wet basis. Each analysis
was carried out in triplicate. If coefficient of variation (CV)
for values were more than 10%, re-analyses were conducted. The
approximate NFE was calculated using the following formula.
