3.5. Genetic stability
Out of 25 primers each for RAPD and ISSR analysis, used for the initial
screening, 12 RAPD and 10 ISSR primers were selected for further analysis based on their resolution and reproducibility (Table 4). In PCRRAPD,
12 primers produced a total of 90 RAPD bands, each ranging from
242 to 3858 bp. The number of bands ranged from 05 to 15 with an
average 7.5 bands per RAPD primer. In ISSR fingerprinting, 10 primers
generated a total of 108 scorable bands ranging from 164 to 4000 bp.
In ISSR fingerprints, the number of bands ranged from 08 to 15 with an
average of 10.8 bands per ISSR primer. The RAPD and ISSR profile
(Fig. 1G & H) of the regenerates and that of the mother plant (P) were
similar in all respects however the occasional appearance of different
bands was not stable across replicates. The monomorphic nature of fingerprints
confirmed the true-to-typeness of in vitro regenerated plantlets;
hence the reliability of regeneration system for genetic restoration
program.
