After mutagenesis, some mutants with few spores on thin
and short mycelia were found. Those colonies were small
and loose, while the mycelia of the parent strain were long
and strong with black spores in the white hyphae, which
formed large and thick colonies. The morphological mutants
were found to produce more lactic acid than the parent strain
and therefore were positive mutants. A morphological mutant
(R. oryzae R1021) with the highest lactic acid productivity,
which formed the largest yellow ring, was obtained
by repeatedly treating with DES, UV and Co60. Starting
with a concentration of 120 g/l corn starch, mutant R1021
produced 79.4 g/l l(+)-lactic acid after 60 h in flasks, 52%
higher than that of the parent strain. The analysis of the fermentation
broth by HPLC demonstrated that 99.05% of lactic
acid produced was l-form. Fig. 4 shows the time course
of lactic acid fermentation by R. oryzae R3017 and R. oryzae
R1021 from glucose in a 5 l stirred tank fermentor. Starting
with a concentration of 88 g/l glucose, the parent strain
R3017 consumed all of the glucose after 30 h and produced
49.1 g/l l(+)-lactic acid, whereas mutant R1021 consumed
all of the glucose after 34 h and produced 69.3 g/l l(+)-lactic
acid. The average productivity of R1021 is 2.04 g/l h, 24.4%
higher than that of the parent strain R3017. The mutant
R1021 was also morphologically different from the parent
strain: the mutant rarely produced hyphae on the selective
plates and the hyphae that did develop were shorter than
those produced by the parent. And in liquid culture the mutant
R1021 formed smaller cell pellets than the parent. The
results also demonstrate that the glass tube selective plate is
very useful to screen the high-lactic acid-producing strain,
and the strain screening strategy of selecting the morphological
mutant is correct.
