Cellularity
We used five different densities of cells prepared using
Papanicolaou staining and PreservCytVR solution:
5 3 105 cells/20 mL, 2.5 3 105 cells/20 mL,
1.25 3 105 cells/20 mL, 0.6 3 105 cells/20 mL,
0.3 3 105 cells/20 mL. The preparation of 5 3 105 cells/
20 mL smeared unevenly because of the high cell density.
Conversely, smears from the 0.6 and 0.3 3 105 cells/
20 mL had very low cell counts and dispersed cells. We
therefore used cells at a density of 1.25 3 105 cells/
20 mL for subsequent experiments (Fig. 1).
