The productionofthermostablexylanasebyanewlyisolated Penicilliumcitrinum xym2wasoptimizedbyone
factoratatime(OFAT)andresponsesurfacemethodology(RSM)approaches.Optimizationofxylanase
productionbyOFATapproachindicatedmediumpH4.0,wheatbran1%(w/v),maltose1.25%(w/v),
di-ammoniumhydrogenphosphate0.09%(w/v),cultivationtemperature30 1C, andincubationtime72has
optimalconditionsforxylanaseproductionof1853IU/ml. Threeeffectivevariables,wheatbranconcentration,
mediumpHandcultivationtemperaturewereoptimized by centralcompositerotatabledesign(CCRD)based
RSM. Theinteractionsbetweenthesevariablescontributedtosignificantincreasedxylanaseproduction.The
RSMoptimizedwheatbranlevel1.5%(w/v),pH(3.5)andtemperature(30 1C) enhancedxylanaseproduction
to2834IU/ml.Theproductionvalueorenzymeproductionwasclosertothemodeldiagnosedvalueof
2845IU/mlandwasabout4foldhigherthaninunoptimizedbasalmedium.Theenzymeshowedpotentialin
saccharification ofsecondgenerationfeedstocksintosugars.
