Pichia pastoris GS115 (his4; Invitrogens) was transformed
with pPIC9K-CtLIP, pJT3, by electroporation with a Bio-
Rad Gene Pulser (Bio-Rads) following the instructions of
the manufacturer. Before transformation, pJT3 was linearized
by digestion with SalI. As a reference, yeast cells were also transformed with the empty vector (pPIC9K) using the
same method. His1 transformants were recovered on MD
plates. To screen for lipase-producing transformants, the
His1 transformants were spotted on MM-rhodamine B agar
and incubated at 30 1C for 3–7 days. Pichia pastoris transformants
were randomly selected and checked for integration
of the CtLIP gene into the genome by PCR.
