Reagents and supplies
1. Invertase solution prepared in advanced from baker’s yeast powder.
Ten gram of dried yeast is dissolved in 40 ml of 0.1 M sodium bicarbonate (pH 4.5) at 40°C for 24 hours. The cell debris is centrifuged off and the supernatant containing the enzyme is stored at 4°C. This solution is diluted 100 times before use.
2. 1% (w/v) Sucrose solution
3. 0.1 M Sodium acetate buffer, pH 3.5, 4.5 and 5.5
4. Benedict’s solution (see Practical I)
