Here we examined for the first time regional differences in neurogenic markers and neural precursor cell characteristics in the hippocampus of the dog. Adult canine hippocampal neurogenesis appears to be strongly dependent on dorsal–ventral location. On the basis of expression of a sequence of neurogenic markers and propensity to formex vivo neurospheres, the canine dorsal dentate gyrus exhibits a higher degree of neurogenesis than the ventral dentate gyrus.
Neural precursors have been isolated from early postnatal canines [19], however, to our knowledge this is the first report to successfully isolate, expand and differentiate neural precursor cells from postmortem adult canine hippocampus. We grew primary hippocampal neurospheres capable of forming secondary neurospheres, quantified neurosphere formation after seeding at clonal density [17], and passaged adherent cells for more than 10 population doublings without evidence of proliferative slowing
