The treatment
potential for inhibitors of GSK-3, including a listing of other
natural product-related structures serving as possible inhibitors
in these disease states, has recently been reviewed.
Using the same basic suite of compounds, it was demonstrated
that indirubins serve as ligands for the “orphan
receptor” known as the aryl hydrocarbon receptor (AhR).
No other natural ligands have yet been identified for AhR,
even though, contrary to earlier beliefs, it has existed for
over 450 million years. Indole-containing compounds, however,
had been suggested as natural ligands for AhR slightly
earlier. Full details of the chemistry involved, and SARs
established using X-ray crystallography and molecular
modeling techniques, have been published
