3.6. Substrate activity
The substrate specificity of amylase indicated that it was active
on a variety of substrates, such as starch, amylopectin, glycogen,
pulullan and maltooligosaccharide, as shown in Table 2. The amylase
from pitaya peel exhibited the highest activity towards starch.
The enzyme also displayed lower amylase activity on amylopectin,
glycogen and pulullan, at 90%, 82% and 70%, respectively, of the
starch-substrate activity. These results indicate that the enzyme
has the capability of cleaving long chains (i.e., alpha-1,6 bonds).
The enzyme partially hydrolysed maltooligosaccharide but did
not achieve any significant (p < 0.05) hydrolysis of a/b-cyclodextrin
(Fig. 4a). Malto-oligosaccharides have two major groups based
on degree of polymerisation (DP): short-length chains and longlength
chains. In the present study, the effect of amylase on the
short-length chain of malto-oligosaccharides with a DP of 4–8
was investigated. Thus, the amylase enzyme, which has a preference
for cleaving long-length chains, displayed lower activity with
malto-oligosaccharides with short-length chains.
