It is evident that the crosstalk between classical genomic and non-genomic actions of estrogen on target genes can finely regulate
gene expression [17]. Accordingly, the relative contributions of genomic and non-genomic actions of estrogen on PSD-95 and
SYP proteins expression were examined by using specific inhibitors of signaling pathways. The results show that 48 h estrogen treatment significantly increased PSD-95 (Fig. 4A) and SYP (Fig. 4B) expression. PI-3K inhibitor, but not MAPK, ROCK, or PKC inhibitors, partially suppressed estrogenic action on both PSD-95 and SYP expression. When used alone, these inhibitors had no effects on basal PSD-95 and SYP expression (data not shown). Therefore, estrogen partially acts through PI-3K signaling to activate PSD-95 and SYP expression. Taken together, with our previous findings that estrogen activates PSD-95 and SYP expression in the classical genomic estrogenic action [4], the possible interactions of classical genomic and non-genomic actions of estrogen-enhance PSD-95 and SYP expressions are evident in SH-SY5Y cells.
