2.5. Analysis of transformation products
The crude extracts obtained from the biotransformation of various
substrates were separated by column chromatography on silica
gel using petroleum ether-ethyl acetate (5:5) to afford 8 and 9,
petroleum ether-ethyl acetate (4:6) to afford 10, dichloromethanemethanol
(15:1) to afford 11 and 12, dichloromethane-methanol
(12:1) to afford 13 and 14, and dichloromethane-methanol (97:3)
to afford 15. Compounds were analyzed by TLC using solvent
system I (chloroform-methanol, 98:2) to separate the transformation
sample of androst-4-en-3,17-dione, solvent system II
(chloroform-methanol, 95:5) to separate the transformation sample
of testosterone, solvent system III (chloroform-methanol, 9:1)
to separate the transformation samples of canrenone and progesterone,
solvent system IV (chloroform-methanol, 85:15) to
separate transformation sample of 16,17-epoxyprogesterone,
and all compounds were visualized by spraying with a solution
of sulfuric acid/ethanol (1:9) followed by heating at 105 ◦C for
5min.
