In conclusion, we have developed a robust approach to
automatically count mammalian cells in images of a cell suspension
in the hemocytometer assembly. The approach takes 2 to 10
times less time than manual counting, leaves a digital trace, yields
the same results as manual counting for the same area but allows
for selection of the most representative areas, and does not impose
a penalty on the number of areas processed, thereby yielding consistent
and more reliable results that are independent of the sample
dilution. A comparison of some parameters for commercially
available cell counters with our approach is presented in
Table S2 of the Supplementary material. Instructions for downloading
and using the macros, along with the code for each macro,
are provided in the Supplementary material as well as at https://
github.com/grishagin/CellCounting.
