Hi All,
I have a quick question. How do you "spot-inoculate" bacteria on an agar plate?
Do you use a toothpick with bacteria (picking up single colony) and stab it into an agar plate? (If it's an aerobic bacteria, I am thinking if they will die of oxygen deficiency.)
Or draw a certain amount (10-50 ul) of liquid bacterial culture with pipette and spot them on the center of an agar plate? (I am thinking the spotted liquid culture will evaporate during the incubation if we do this way, and they can't get enough nutrient just being on the surface of the agar plate)
I am working with bacteria, not fungi. Do you have any suggestion? Thanks a lot in advance!
