Upon subsequent magnetic separations of cells over a MACS® Column (LS Column) that
was placed in a magnetic field of a MACS Separator, the magnetically labeled non-NK cells (T cells, B
cells and other debris) were retained within the column, while the unlabeled NK cells ran through. The
fraction of NK cells was decanted into sterile Universal tubes and centrifuged (400 g for 10 minutes). The
pellet was resuspended in RPMI 1640, viable cell number determined and cells diluted with medium to
give the appropriate concentration for experiments.
