In this experiment, HPLC is used to determine caffeine content in 2 brands of tea beverages, oishi and fuji brand. In HPLC, each tea sample is injected into a mobile liquid phase and it passes along a stationary phase. After the sample passes through the column, it is detected by ultraviolet absorption(used272.0 nm). The sample and the mobile phase are collected as waste and the absorption spectrum is outputted as a chromatogram.
For the result,the oishi sample give peak area 1088596 µv•sec, retention time 3.891 minutes and have caffeine 45.14 mg. Then the fuji sample give peak area 1952883 µv•sec, retention time 3.883 minutes and have caffeine 91.30 mg. So the tea brand which have most of caffeine is Fuji brand and high caffeine can hace bad effect to human body such as a headache, heartburn and it can lead to bone thinning. And from the graph of caffeine standard solution is tell that there are some error in this experiment because the line is a little bit not in a linear line.
