3.2. Sensitivity of real-time RT-PCR
To determine the sensitivity of real-time RT-PCR, 10-fold serially diluted
CMNV plasmid which containing the target sequence was used as
a template for preparing the standard. The TaqMan real-time RT-PCR
gave linear curves for 108 to 1 copies of pCMNV (Fig. 2a). The sensitivity
per reaction of the assaywas as high as 1 copy detection of pCMNVwith
correlation coefficient (R2) of 0.9915 and a slope of 3.002 (Fig. 2b).
These values were used to calculate the number of CMNV RNA copies
in shrimp samples.
