1. Inoculate two tubes of glucose oxidation-fermentation agar with each of bacterial cultures provided. Stab the medium down to the bottom of the tube.
2. Cover one tube of each pair of inoculated media with a thin layer of sterile mineral oil.
3. Incubate all tubes at 37 oC for 24 hrs.
4. After incubation, examine each tube for the production of acid (yellow color).
5. Examine each culture for the presence of turbidity or cloudiness. This medium will turn cloudy with a motile culture because of the movement of organisms through the soft agar. The medium remains clear with non-motile culture.
