The bacterioMatch II two-hybrid system (stratagene) was used to detect protein-protein interactions. The plasmid pairs were used to cotransform the XL1-Blue MRF' reporter strain on M9 salt agar without 3-amino-1,2,4(3-AT).Colonies were then restreaked on M9 salt agar containing 5 mM 3-AT at 37'C for 24 hour for the first detection of interactions.
For confirmation, the colonies were cultured on dual-selective medium containing 5 mM 3AT and streptomycin as described in the manual.
