Undiluted milk samples (0.5 mL) were saponified by the addition of 10 mM
0.5% (wt/vol) sodium dodecyl sulfate (JT Baker), 50 mL 60% (wt/vol) potassium
hydroxide in water, and 1 mL ethanol followed by vigorous shaking for 30s.
Hydrolyzed samples were then extracted three times with hexane, evaporated
under nitrogen, and resuspended in 50 mL dichloromethane and 450 mL HPLC
mobile phase (75% acetonitrile, 15% methanol, 10% dichloromethane, 1.54 g/L
ammonium acetate, and 1.0 mL/L triethylamine).
