As shown in Fig. 1, the Basmati rice strain was crossed with the
Myanmar rice cultivar Manawthukha, using Manawthukha as the
female parent and Basmati as the male parent, to develop the F1
generation. The F1 plants were backcrossed to Manawthukha using
mixed pollen to generate the BC1F1 generation. Consecutive backcrossing
was conducted until the BC4F1 generation was produced.
MAS was carried out using 200 individuals in each generation of
backcrossing and BC4F2 population. The plants that carried heterozygous
alleles of certain markers were selected and crossed with
Manawthukha. The selected plants in BC4F1 were self-pollinated to
generate the BC4F2 population. Twelve BC4F2 lines possessing
homozygous Basmati alleles were successfully developed.
