‘Simulated gastric fluid TS’ is formally referred to as ‘Gastric fluid, simulated TS (Test solution)’. It is a commonly used dissolution media that is described in United States Pharmacopeia 33-28NF (2010) and European Pharmacopeia 7.0 (2010).
Simulated gastric fluid TS is an artificial dissolution medium that is intended to represent stomach acid. It is prepared by dissolving 2.0 g of sodium chloride and 3.2 g of purified pepsin (derived from porcine stomach mucosa, with an activity of 800 to 2500 units per mg of protein), in 7.0 mL of hydrochloric acid and water up to 1000 mL. This test solution has a pH of about 1.2. To simplify its preparation, if hard gelatin capsules are not being tested, pepsin can often be omitted from this official media and has been described by Galia et al[1].
